If the BIOS of the system allows for booting from and reading of GPT layouts on a hard drive, you should be able to stick a >2TB drive into it and use it with a GPT layout. MBR uses 32 bits for LBA and is limited to 2TB drives, whereas GPT uses 64 bits, allowing for much higher capacities.
Some older machines may choke on larger drives. Your mileage may vary, though. As for OS support, both 32 and 64-bit Linux, the BSDs, OS X, and 64-bit Winbloze all boot from GPT drives.
